PREVENTIVE EFFECTS OF TOCOTRIENOL-ENRICHED MIXED FRACTION ON PLAQUE FORMATION AND STABILITY IN EARLY AND ESTABLISHED ATHEROSCLEROSIS

Atherosclerosis is the underlying pathology for cardiovascular disease. The preventive effects of tocotrienol-enriched mixed fraction (TEMF) on atherogenesis remains unclear. Objective: To investigate TEMF supplementation early and established atherosclerosis. Methods: Twenty New Zealand white rabbits were divided into (n=10) placebo groups. Treatments given by oral gavage 8 weeks followed 1% high cholesterol diet (HCD) another two (to induce atherosclerosis) or eight (established atherosclerosis). At end study, aorta was dissected, stained with Sudan IV, qualitatively analysed atherosclerotic lesion. Immunohistochemistry performed to detect expression interleukin-6 (IL-6), CRP, nuclear factor kappa beta, E-selectin, intercellular adhesion molecule-1 (ICAM-1), vascular cell protein-1 (VCAM-1), smooth muscle actin (SMA) matrix metalloproteinase-12 (MMP-12). Results: In both atherosclerosis groups, there a significant reduction lesions in compared placebo. Atherogenic biomarkers; IL-6, SMA, MMP-12 reduced (p<0.05). Neutral seen group. setting decreased formation reduces endothelial inflammation lesions, while increasing plaque stability.
